Can diet plus inositol or metformin tame insulin resistance in PCOS?
NCT ID NCT07770711
First seen Aug 18, 2026 · Last updated Aug 18, 2026
Summary
This trial tests whether adding inositol or metformin to either a Mediterranean or low-carbohydrate diet improves insulin resistance in obese women with polycystic ovary syndrome (PCOS). About 90 to 102 participants will be randomly assigned to one of six groups and followed for two months. The main goal is to see if insulin resistance, measured by HOMA-IR, changes from the start to the end of the study. Secondary measures include changes in body weight, body composition, and metabolic markers.
What this could mean
Our plain-language read of the trial. This is informational only — not medical advice or a prediction.
- Active substance
- Metformin (drug) and myo-inositol (dietary supplement), each combined with either a Mediterranean or low-carbohydrate diet, compared against folic acid as an active control.
- What this could lead to
- If successful, this could identify a more effective diet-and-supplement combination to improve insulin resistance and metabolic health in women with PCOS, potentially guiding future treatment recommendations.
- What could go wrong
- This is a small, short-term (2-month) trial, so results may not be durable or generalizable. Gastrointestinal side effects from metformin are possible, and the open-label design may introduce bias.
