Could antidepressants finally silence chronic cough?

NCT ID NCT05110144

First seen Jun 27, 2026 · Last updated Jun 27, 2026

Summary

This study tests whether two common antidepressants, duloxetine and amitriptyline, can reduce coughing in people with refractory chronic cough—a cough that persists despite standard treatments. About 50 adults will receive either a low or high dose of one drug or a placebo for 4 weeks. Researchers will measure cough frequency using a special monitor to see if these drugs offer relief.

What this could lead to

If it works, this could point toward a new treatment option for people with chronic cough that doesn't respond to other therapies.

What could go wrong

This is an early phase 2 trial with only 50 people, so results may not apply to everyone. The drugs can cause side effects like drowsiness, dry mouth, or nausea.
