Immunotherapy before Chemo-Radiation shows promise for Hard-to-Treat cancers
NCT ID NCT04165772
First seen Jun 25, 2026 · Last updated Jun 27, 2026 · Updated 2 times
Summary
This study is testing whether the immunotherapy drug dostarlimab (TSR-042), followed by standard chemoradiation and surgery, can effectively treat advanced mismatch repair deficient (dMMR) solid tumors. About 212 adults with locally advanced dMMR cancers will receive the drug intravenously every 3 weeks, then chemoradiation, and finally surgery. The main goal is to see if the cancer completely disappears or shrinks significantly within 12 months.
What this could mean
Our plain-language read of the trial. This is informational only — not medical advice or a prediction.
- Active substance
- Dostarlimab (TSR-042), capecitabine or 5-FU, and radiation therapy
- What this could lead to
- If successful, this approach could offer a way to shrink or eliminate advanced dMMR tumors before surgery, potentially reducing the need for more aggressive treatments.
- What could go wrong
- This is a phase 2 trial, so results are still early. The combination may cause side effects from chemotherapy, radiation, or immunotherapy, and not all patients may respond.
