Richter transformation
MONDO:0002083Transformation of chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL) into a more aggressive type of lymphoma, usually diffuse large B-cell lymphoma (immunoblastic or centroblastic variant). Occasional cases of transformation to Hodgkin's lymphoma have also been described, particularly in patients treated with purine nucleotide analogs. Molecular genetic studies suggest that in approximately half of the cases, the lymphoma is clonally related to the underlying CLL/SLL, whereas in the remaining cases the lymphoma probably represents a secondary, unrelated neoplasm.
Also known as: Richter syndrome, Richter's syndrome, Richter's transformation, chronic lymphocytic leukemia with Richter transformation, chronic lymphocytic leukemia/small lymphocytic lymphoma with Richter transformation

New hope for richter transformation: targeted drug combo heads to phase III
Disease control Not yet recruitingThis study tests whether adding the targeted drug pirtobrutinib to standard chemotherapy (R-CHOP) works better than chemo alone for people with Richter transformation, a rare and aggressive lymphoma that develops from chronic lymphocytic leukemia. About 102 adults will be randoml…

New cell therapy aims to wipe out lingering lymphoma before it returns
Disease control Not yet recruitingThis early-phase trial tests a personalized cell therapy (rapcabtagene autoleucel) in 20 adults with large B-cell lymphoma who still have traces of cancer after initial treatment. The goal is to see if the therapy can make those traces disappear, potentially lowering the risk of …

Which Pre-CAR-T chemo is safer? new trial aims to find out
Disease control Not yet recruitingThis trial tests whether bendamustine causes fewer severe side effects than the standard fludarabine/cyclophosphamide combination when given before CAR-T cell therapy for B-cell lymphoma. About 92 people will be randomly assigned to one of the two chemotherapy regimens. The main …

Smart radiation: new trial aims to zap lingering lymphoma cells
Disease control Not yet recruitingThis study tests whether using a blood test (MRD) after two cycles of chemoimmunotherapy can guide the use of low-dose radiation in patients with diffuse large B-cell lymphoma. Patients with detectable cancer DNA in their blood may receive targeted radiation to remaining spots on…

Experimental CAR-T therapy takes on tough lymphoma
Disease control Not yet recruitingThis early-phase trial is testing a new type of immune cell therapy called CD19X CAR-T for people with large B-cell lymphoma that has come back or not responded to other treatments. Only 3 participants will receive the therapy after a short course of chemotherapy to prepare their…

New antibody combo targets tough lymphoma in early trial
Disease control Not yet recruitingThis study tests a new drug called EMB-07, which is a bispecific antibody that helps the immune system attack cancer cells. It is for people with aggressive B-cell non-Hodgkin lymphoma, either newly diagnosed or whose cancer has come back. The trial will check safety, side effect…
