Drug showdown: which pill tames cortisol better?

NCT ID NCT07268222

First seen Jun 27, 2026 · Last updated Jun 27, 2026

Summary

This study compares two medications, metyrapone and osilodrostat, in 150 adults with mild autonomous cortisol secretion (MACS). Participants receive one of the drugs, surgery, or watchful waiting. The goal is to see which drug better lowers cortisol and improves related health issues like high blood pressure and blood sugar over 6 months.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

metyrapone and osilodrostat (oral tablets)

What this could lead to

If successful, this trial could show which drug better controls cortisol levels and improves blood pressure, weight, and blood sugar in people with mild autonomous cortisol secretion.

What could go wrong

This is a relatively small Phase 4 trial, so results may not apply to everyone. Both drugs can cause side effects like nausea or low cortisol, and the study does not test a cure.
