New combo therapy aims to shrink breast tumors before surgery

NCT ID NCT06650748

First seen Jun 24, 2026 · Last updated Jun 27, 2026 · Updated 1 time

Summary

This phase 2 trial is testing whether adding the targeted drug dalpiciclib to standard hormone therapy (letrozole) before surgery can improve outcomes for women with HR+/HER2- early-stage breast cancer. The study enrolls 100 women aged 18-70 whose tumors have a high genetic risk score and show a poor early response to hormone therapy. The goal is to see if this combination leads to a complete or near-complete disappearance of the tumor by the time of surgery.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

Dalpiciclib (a CDK4/6 inhibitor) and letrozole (an aromatase inhibitor)

What this could lead to

If successful, this could lead to a more personalized treatment approach for early-stage breast cancer, potentially improving response rates before surgery.

What could go wrong

This is a small, early-phase trial (100 participants) at a single center, so results may not apply broadly. Side effects from the drug combination, such as low blood cell counts or fatigue, are possible.
