Triple threat: new combo therapy aims to tackle rare lung disease

NCT ID NCT07266519

First seen Jun 27, 2026 · Last updated Jun 27, 2026

Summary

This phase 2 trial tests whether a triple combination of three drugs—sotatercept, an endothelin receptor antagonist, and a phosphodiesterase-5 inhibitor—can improve blood flow in the lungs of people newly diagnosed with pulmonary arterial hypertension (PAH). The study will enroll 25 treatment-naïve adults and measure changes in pulmonary vascular resistance after 24 weeks. It is an open-label, single-arm study, meaning everyone gets the same treatment and no placebo is used.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

sotatercept combined with an endothelin receptor antagonist and a phosphodiesterase-5 inhibitor

What this could lead to

If successful, this triple-drug combination could become a new standard first treatment for pulmonary arterial hypertension, potentially improving blood flow and symptoms better than current options.

What could go wrong

This is a small, early-phase study with only 25 people and no comparison group. Results may not apply to all patients, and side effects from combining three drugs are unknown.
