Experimental drug combo targets Hard-to-Treat HER2 cancers

NCT ID NCT07137416

First seen Jan 06, 2026 · Last updated Jun 19, 2026 · Updated 29 times

Summary

This early-phase trial tests the safety and best dose of a new drug combination—pidnarulex plus trastuzumab deruxtecan—in people with advanced breast cancer and other solid tumors that have the HER2 protein. The study enrolls 36 adults whose cancer has spread or cannot be surgically removed. Pidnarulex is an enzyme blocker that kills cancer cells, while trastuzumab deruxtecan is a targeted therapy that delivers chemotherapy directly to HER2-positive cells. The goal is to find a tolerable dose and see early signs of tumor shrinkage.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

pidnarulex and trastuzumab deruxtecan

What this could lead to

If this combination proves safe and effective, it could offer a new treatment option for people with advanced HER2-positive or HER2-low breast cancer and other solid tumors.

What could go wrong

This is a very early Phase 1 trial with only 36 participants. The main goal is safety and dosing, not proof of effectiveness. Side effects from the drug combination could be significant, and many early-stage cancer trials do not lead to approved treatments.
