NCT ID NCT05786924
First seen Nov 03, 2025 · Last updated Jun 23, 2026 · Updated 42 times
Summary
This study tests an experimental oral drug called S241656 (BDTX-4933) in people with advanced solid tumors that have certain genetic changes (KRAS, BRAF, or other RAS/MAPK mutations). The trial has two parts: first, finding a safe dose, then testing how well it works alone or with other cancer drugs. About 554 adults with lung, colorectal, pancreatic, or other cancers will take the drug daily in 28-day cycles. The goal is to see if the drug can shrink tumors or slow disease progression.

What this could mean
Our plain-language read of the trial. This is informational only — not medical advice or a prediction.
Active substance
S241656 (also called BDTX-4933), an oral RAF inhibitor
What this could lead to
If successful, this could provide a new treatment option for people with advanced cancers that have specific genetic mutations (like KRAS or BRAF) and have not responded to other therapies.
What could go wrong
This is an early-phase trial (Phase 1/2) with a small number of participants, so the drug may not work as hoped or could cause unexpected side effects. It is also testing many different cancer types and combinations, making it harder to prove benefit for any single group.
Conditions
The condition(s) this trial relates to.
As listed by the trial registrant
The condition terms exactly as the trial's registrant entered them.
