Promising new combo aims to shrink tumors faster in kids with tough hodgkin lymphoma
NCT ID NCT01920932
First seen Jun 26, 2026 · Last updated Jun 27, 2026 · Updated 1 time
Summary
This phase 2 trial tests whether replacing a standard chemo drug with Adcetris (brentuximab vedotin) can improve early tumor shrinkage in children with high-risk Hodgkin lymphoma. About 77 participants will receive two cycles of the new combo, followed by more chemo and possibly radiation. The goal is to see if more kids achieve a complete response early, potentially reducing the need for radiation.
What this could mean
Our plain-language read of the trial. This is informational only, not medical advice or a prediction.
- Active substance
- brentuximab vedotin (Adcetris) plus chemotherapy (etoposide, prednisone, doxorubicin, cyclophosphamide, dacarbazine) and radiation therapy
- What this could lead to
- If successful, this approach could improve early cancer clearance and reduce the need for radiation in children with high-risk Hodgkin lymphoma.
- What could go wrong
- This is a small, early-phase trial (77 participants) with no control group. Results may not generalize, and side effects from the drug combination could be significant.

Inclusion Criteria: * Histologically confirmed, previously untreated CD30+ classical Hodgkin Lymphoma (HL). (Participants receiving limited emergent radiation therapy (RT) or steroid therapy - maximum of 7 days - because of cardiopulmonary decompensation or spinal cord compression will be eligible for protocol enrollment). * Age ≤ 18 years at the time of diagnosis (i.e., participants are eligible until their 19th birthday). * Ann Arbor stage IIB, IIIB, IVA, or IVB. * Adequate renal function based on GFR ≥ 70 ml/min/1.73m\^2 or serum creatinine adjusted for age and gender. * Adequate hepatic function (total bilirubin \< 1.5 x ULN for age, and SGOT/SGPT \< 2.5 x ULN for age). * Female participant who is post-menarchal must have a negative urine or serum pregnancy test. * Female or male participant of reproductive potential must agree to use an effective contraceptive method throughout duration of study treatment. Exclusion Criteria: * CD30 negative HL. * Has received prior therapy for Hodgkin lymphoma, except as noted above. * Inadequate organ function as described above. * Inability or unwillingness of research participant or legal guardian / representative to give written informed consent.
