Bone drug may stop Paget's before it starts in at-risk families

NCT ID NCT03859895

First seen Jun 27, 2026 · Last updated Jun 27, 2026

Summary

This study follows people who carry a gene mutation that puts them at high risk for Paget's disease of the bone. They previously received either zoledronic acid or a placebo in the ZiPP trial. Now researchers will track them for another 5 years to see if the drug can prevent or delay bone lesions and symptoms. The goal is to learn whether early treatment can stop the disease from ever causing problems.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

zoledronic acid (a bisphosphonate drug)

What this could lead to

If it works, this could point toward a way to prevent Paget's disease in people who carry the high-risk gene mutation.

What could go wrong

This is an extension of an earlier trial, and it is still unclear whether preventing bone lesions will actually prevent symptoms. The study is small and focused on a specific genetic group, so results may not apply to everyone.
