Expanded access (not trials)

Expanded access
Not a trial. This treatment can be requested outside a study, case by case, for people who qualify.
Expanded access (paused)
Not a trial. The treatment can normally be requested outside a study, but is unavailable right now.
Expanded access (ended)
Not a trial. The treatment could once be requested outside a study, but no longer can.

Summary

Researchers are testing an experimental drug called IBI343 in people with advanced pancreatic cancer that has worsened after at least two rounds of chemotherapy. The trial enrolls about 200 adults whose tumors carry a protein called CLDN18.2. Participants receive either IBI343 or a placebo, both given by intravenous infusion every three weeks along with supportive care. The main question is whether IBI343 helps people live longer than placebo.

What this could mean

Our plain-language read of the trial. This is informational only, not medical advice or a prediction.

Active substance
IBI343, an experimental antibody-drug conjugate that targets the CLDN18.2 protein on cancer cells
What this could lead to
If IBI343 works, it could give people with advanced pancreatic cancer a new treatment option after standard chemotherapy stops helping. It could also confirm that targeting CLDN18.2 is a useful strategy for this hard-to-treat cancer.
What could go wrong
Pancreatic cancer is aggressive and often resists treatment, so IBI343 may not extend survival more than supportive care alone. The trial enrolls about 200 people, and results may not apply to everyone with this cancer. Antibody-drug conjugates can cause side effects such as nausea, fatigue, and low blood counts.

Inclusion Criteria: 1. Sign the written informed consent form (ICF) and be willing and able to comply with the visits and related procedures stipulated in the plan. 2. Histologically confirmed unresectable locally advanced, or metastatic pancreatic cancer. 3. Have received and progression after at least two systemic therapies(must including a fluorouracil-based and a gemcitabine-based therapy). 4. ECOG PS score of 0 or 2. 5. Adequate bone marrow and organ function 6. Confirmed as CLDN18.2 positive. Exclusion Criteria: 1. Participation in another interventional study, except observational or post-intervention follow-up. 2. Prior treatment with topoisomerase inhibitor-based ADC. 3. Has received the last dose of an anti-cancer therapy within 2 weeks or 5 half-lives (whichever is shorter) prior to the first dose of study treatment. 4. Plans to receive other anti-tumor treatments during treatment with the study drug (palliative radiotherapy for symptomatic (e.g., pain) relief that does not affect response assessment is allowed) . 5. Symptomatic CNS metastasis; asymptomatic brain metastases may be allowed with specific criteria. 6. History of other primary malignancies, except cured or low-risk of recurrence.
