New combo therapy aims to heal Radiation-Damaged jaw bone

NCT ID NCT06055257

First seen Jun 26, 2026 · Last updated Jun 26, 2026

Summary

Radiation for head and neck cancer can sometimes cause a serious side effect: dead bone in the jaw (osteoradionecrosis). This pilot study tests whether adding a drug cocktail called PENTOCLO to standard hyperbaric oxygen therapy helps more than oxygen alone. Twenty-four adults with jaw bone damage will be randomly assigned to one of the two treatments. The main goal is to see if a larger trial is possible, but researchers will also track pain, side effects, and whether surgery is needed.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

hyperbaric oxygen therapy and a modified PENTOCLO regimen (pentoxifylline, tocopherol, and clodronate, preceded by antibiotics and an antifungal)

What this could lead to

If this pilot succeeds, it could point toward a more effective treatment for radiation-induced jaw bone damage, potentially reducing pain and the need for surgery.

What could go wrong

This is a very small pilot study (24 people) focused on feasibility, not proof of effectiveness. The combination therapy is complex and requires many clinic visits and daily pills, which may be hard for patients to complete.
