Experimental pill plus chemo takes aim at Hard-to-Treat brain lymphoma

NCT ID NCT07608731

First seen Jun 26, 2026 · Last updated Jun 27, 2026 · Updated 1 time

Summary

This early-phase trial tests a new oral drug called golcadomide combined with standard chemotherapy drugs (pemetrexed, rituximab, and dexamethasone) in 18 adults with large B-cell lymphoma that has spread to the brain or central nervous system and has either returned or is not responding to current treatment. The main goals are to check the safety of the combination and find the best dose. Participants will receive up to six 21-day cycles of treatment and be followed for about 2.5 years.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance
Golcadomide (an experimental drug taken orally) combined with pemetrexed, rituximab, and dexamethasone (standard chemotherapy drugs)
What this could lead to
If this works, it could point toward a new treatment option for people with CNS lymphoma that has come back or stopped responding to therapy.
What could go wrong
This is a very early Phase 1 trial with only 18 participants, so it is primarily testing safety and dosing—not yet proving effectiveness. The drug combination may cause side effects or fail to shrink tumors.
