New drug cocktail aims to shrink breast tumors before surgery

NCT ID NCT07492394

First seen Jun 25, 2026 · Last updated Jun 27, 2026 · Updated 2 times

Summary

This Phase 2 trial tests whether adding entinostat to a two-drug regimen (dalpiciclib and letrozole) before surgery can better shrink tumors in women with HR-positive, HER2-negative early breast cancer. Sixty participants will receive either the three-drug combo or the standard two-drug combo for six cycles before surgery. The study measures how many patients have a complete or partial response, and how many achieve a pathological complete response (no cancer left in breast or lymph nodes).

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance
Dalpiciclib, entinostat, and letrozole
What this could lead to
If this works, it could point toward a more effective pre-surgery treatment for HR+/HER2- early breast cancer, potentially improving tumor shrinkage and surgical outcomes.
What could go wrong
This is a small, early-phase (Phase 2) study with only 60 participants, so results may not apply to all patients. Adding entinostat may increase side effects, and the benefit over standard therapy is uncertain.
