Could a common arthritis drug ease knee pain? new trial investigates

NCT ID NCT07161336

First seen Nov 01, 2025 · Last updated Jun 22, 2026 · Updated 29 times

Summary

This phase 2 trial tests whether methotrexate, a drug used for rheumatoid arthritis, can reduce pain and improve function in people with knee osteoarthritis who also have joint inflammation (synovitis) seen on ultrasound. Seventy adults aged 40-70 will receive either methotrexate or a placebo for 6 months, and researchers will measure changes in pain, stiffness, and daily activities. The goal is to see if targeting inflammation can offer relief for a condition with limited treatment options.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

Methotrexate

What this could lead to

If it works, this could point toward a new treatment option for knee osteoarthritis patients with joint inflammation, potentially easing pain and improving mobility.

What could go wrong

This is a small, early-phase trial with only 70 participants, so results may not apply to everyone. Methotrexate can cause side effects like nausea or liver issues, and it may not prove more effective than placebo.
