New drug shows promise in shrinking aggressive breast tumors before surgery
NCT ID NCT05900206
First seen Oct 31, 2025 · Last updated Jun 23, 2026 · Updated 34 times
Summary
This study compares a newer targeted drug, trastuzumab deruxtecan (T-DXd), with standard chemotherapy and targeted therapy given before surgery for HER2-positive breast cancer. About 370 adults with large or lymph node-positive tumors will receive either T-DXd or standard treatment for three cycles, then further treatment based on tumor biology. The main goal is to see if T-DXd leads to a higher rate of complete tumor disappearance at surgery.

What this could mean
Our plain-language read of the trial. This is informational only — not medical advice or a prediction.
Active substance
trastuzumab deruxtecan (T-DXd, Enhertu)
What this could lead to
If successful, this could lead to a more effective pre-surgery treatment for HER2-positive breast cancer, potentially improving cure rates and reducing the need for chemotherapy.
What could go wrong
This is a phase 2 trial with 370 participants, so results are preliminary. The new drug may not be more effective than standard care, and side effects like lung inflammation or heart problems are possible.
Conditions
