New combo shot aims to reignite immune attack on tough melanoma

NCT ID NCT04796194

First seen Jun 25, 2026 · Last updated Jun 27, 2026 · Updated 1 time

Summary

This phase 2 trial tested a combination of an injected drug (LTX-315) and an intravenous immunotherapy (pembrolizumab) in 23 people with advanced melanoma that no longer responded to standard PD-1/PD-L1 inhibitors. The goal was to see if the combination could shrink tumors or slow the disease. Participants had tumors that could be injected directly, such as those under the skin or in lymph nodes.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

LTX-315 (injected into tumors) combined with pembrolizumab (IV infusion)

What this could lead to

If successful, this combination could offer a new treatment option for advanced melanoma patients whose cancer no longer responds to standard immunotherapy.

What could go wrong

This is a small, early-phase study with only 23 participants, so results may not apply broadly. The treatment involves injections into tumors and may cause side effects like pain, swelling, or immune-related reactions.
