New drug combo targets Hard-to-Treat cancers in early trial

NCT ID NCT04418167

First seen Nov 01, 2025 · Last updated Jun 23, 2026 · Updated 39 times

Summary

This early-stage study tests a new drug called JSI-1187, either alone or combined with dabrafenib, in people with advanced solid tumors that have certain genetic changes (MAPK pathway mutations). The main goal is to check safety and tolerability, with a secondary look at whether the tumors shrink. About 71 adults with tumors that have not responded to standard treatments are being enrolled. Because this is a Phase 1 trial, it is too early to know if the drug will work, and the study is currently on hold.

What this could mean

Our plain-language read of the trial. This is informational only — not medical advice or a prediction.

Active substance

JSI-1187 (an ERK1/2 inhibitor) and dabrafenib (a targeted cancer drug)

What this could lead to

If successful, this could point toward a new treatment option for people with advanced solid tumors that have specific genetic mutations.

What could go wrong

This is a very early Phase 1 trial with only 71 participants, so it is primarily testing safety, not effectiveness. The study is currently suspended, and many early cancer drugs fail to show enough benefit to move forward.

Conditions

The condition(s) this trial relates to.
